Development of One Cell Stage Mouse Zygotes in Simple(EBSS) and Complex(Hanm´s F-10)Media and The Effect of Fetal Cords Serum and Protein

Abstract

|
|
One cell stage mouse zygotes were cultured in Ham´s F-10 alone, Ham´s F-10 with 10% cord serum (C.S.), Ham´s F-10 with 5 ㎎/㎖ Bovine Serum Albumin (BSA), Earle´s balanced salts solution (EBSS) alone, EBSS with 10% C.S., EBSS with 5 ㎎/㎖ BSA.
Development was assessed by the percent of embryos developing to the 2-cell stage and on to the blastocyst stage.
Development to the 2-cell was not different by the different medium, but development to the blastocyst was significantly different. Development to the blastocyst stage was significantly higher in Ham´s F-10 than in EBSS (p<0.012) and higher in Ham´s F-10 with 10% CS or 5㎎/㎖ BSA than Ham´s F-10 (p<0.08, p<0.113) and significantly higher in EBSS with 10% CS or 5㎎/㎖ BSA than EBSS (p<0.001).
In our results, cord serum and portein promot one cell stage mouse zygote development to the blastocyst stage.
